Summary of experimentsThe region from -2844 to -2720 is required for expression in the a6.5 lineage. This region is hence called the a-element.
This region contains 2 Ets boxes and 7 GATA boxes, of which only the last 3 (around the Ets boxes) are critical.
Removal of 1 Ets box is results in lowered expression, while the removal of both abolished expression completely.
Both maternal Ets1/2 and GATAa are necessary for expression of the a-element. They act cooperatively.
Expression driven by the a-element is induced by FGF9/16/20 (Bertrand 2003)

The region from -2720 to -2436 is require for expression in the A-line (Bertrand 2003)

The region from -2009 to -1574 is required for expression in the B-line (Bertrand 2003)

Ci-macho1 binds to 5'-GCCCCCCGCTG-3', the terminal Gs and the central CCC being critical for binding in vitro (Yagi 2004)
Despite the presence of possible Ci-macho1 binding sites the regulatory effects reported by Yagi et al. could be indirect.

Severely reduced expression in the absence of Ci-Rga (could be indirect) (Hamada 2005).
